The role of lactate dehydrogenase in the progression of breast cancer cell lines MCF-7 & MDA-MB231 and its inhibition by oxamate

Introduction & Aim: Higher rate of glycolysis has been long observed in cancer cells, as a vital enzyme in glycolysis, lactate dehydrogenase (LDH) has been shown with great potential as an anti-cancer target. Here, we investigated the effects of oxamate, one classic inhibitor of LDH-A in MCF-7 & MDA-MB231 cell lines.Methods: In this study, after culturing cancer cells and adding oxamate to cells, the proliferation of cells and the activity of the enzyme lactate dehydrogenase as the main marker of glycolysis pathway was determined by MTT assay and LDH activity assay kit respectively.Results : The results showed that oxamate significantly suppressed the proliferation of MCF-7 & MDA-MB231 cells. Furthermore, in the present study, we found that the power of oxamate as an inhibitor of LDH-A in MDA-MB231 greater than to MCF-7.Conclusion: Collectively, these results suggest that LDH-A may serve as a promising therapeutic target for breast cancer treatment.
