Increasing production of clavulanic acid and changing of consumption carbohydrates patterns in Streptomyces clavuligerus by ultraviolet mutagenesis

Streptomyces is a soil-born, aerobic Gram-positive bacteria from Actinomaystal order. Different species of the Streptomyces produce a wide range of antibiotics. Streptomyces clavuligerus, a famous actinomy-cetes, is capable of producing more than 20 kinds of antibiotics, especially commercial antibiotic, clavu-lanic acid (CA). CA is a beta-lactamase inhibitor and prevent beta-lactam antibiotics from inactivation. The combination of CA with amoxicillin (co-amoxiclave) is one of the most commonly used antibiotics in the world. One of the most important issues in biotechnology, is to enhance strain optimization ac-tivities, to increase crop productivity with high economic efficiency. For this purpose, various methods including chemical or physical mutagenesis can be used. Among the most important factors in physical mutagens is ultraviolet radiation from 200 to 300 nm to increase CA production. In this project, Strep-tomyces clavuligerus DSMZ 738 were used to increase the producing of CA by mutation with UV beams with a wavelength of 250 nm and the concentration of CA was measured by HPLC. The resulting mutants produced 92% CA than that of the parent strain. The production of CA in the ISP4 medium was %83 less. However, in all media studied, CA production by mutants was greater than the parent strain. The results show that UV radiation affects glucose, arabinose, sucrose and xylose. the mutants were able to break down the sugars more rapidly. On the other hand, there was no change in glucose, inositol, mannose,rhamnose, raffinose and cellulose
