Tissue engineering is a field for the repair of damaged tissues, which can facilitate the substitution at the damaged sites by engineered tissues with suitable characteristics. In this research, silk fibroin / chitosan nanofibers prepared with electrospinning technique as a tissue scaffold. Silk fibroin extracted from silkworm cocoons in the first step. Then chitosan and silk fibroin polymer dissolved with weight ratio 7:3 in a mixed solvent system of TFA/DCM. The electrospun SF/CS blend nanofibers were collected on a target drum. The voltage in range of 12-18 kV were applied by a high voltage power supply. The beadless nanofibers scaffold were assayed by SEM imaging for determining morphology and average diameter of nanofibers. For degradation invitro assay, Phosphate-buffered saline was used during 84 days. SEM images results showed that the nanofibers diameter were in the range of 80-170 nm. During invitrio assay, more than 95% of total scaffold weight remained. FTIR spectroscopy showed changes on nanofibers surface. These polymers contain suitable functional groups that prepare a lot of domains for cells attaching without surface modification. Slow degradation and hydrophilic surface promoting cell adhesion, good biocompatibility and antibacterial activity of chitosan give suitable features to this scaffold.
