چکیده مقاله:

Dermaneh dashti (Artemisia sieberi Besser) is a perennial plant belonging to Asteraceae family. It is a very valuable medicinal effects such as antioxidant, antifungal, antimalarial and antidiabetic. Propagation of this plant was happened with prolonged dormancy seeds that needs to spend more than one year on the stock of mother which has been effective in reducing the population, so propagation of Artemisia sieberi with tissue culture is most important. Elhag et al (1991) were reported that MS medium containing 1 mg/l NAA and 1/0 mg/l Kinetin is effective in callus induction in Artemisia annua.MATHERIAL AND METHODS: The first seeds were washed with distilled water and soaking in alcohol (96%) for 5 minutes. Then they were disinfected by 25% sodium hypochlorite for 5 min and washed with distilled water, then they cultured in half Murashig and Skoog medium. After 90 days were prepared nodals from explants of seeds and were cultured in DKW medium with different levels of BA (0, 0/1, 3/0 mg/l), NAA (0, 2/0, 5/0 mg/l) and GA3 (0 ,2 ,4 mg/l) and after 60 days in explans were determined the number of shoots and roots and callus. This experiment was performed in form of randomized completely factorial design with three replicates and two explants in each repetition. Data analysis was performed with SAS and comparing the averages with Duncans multiple range test.RESULTS AND DISCUSSION: Results showed that medium containing 3/0 mg/l BA, 5/0 mg/l NAA and 2 mg/l GA3was effective in increasing the number of shoot (29/50 per each explants) that showed a significant differences at level of 5% with control. GA3 with CK was increased the proliferation that similar results of Sujatha et al (2007) were obtained that they were reported application of 4/44 μm BA and 2/32 μm Kinetin in Artemisia vulgaris showed maximum shooting (99/8 percent) and the highest rooting (98/2 percent) in medium containing 8/65 μm IAA. NAA caused rooting and GA3 is effective on elongation of roots. Similar results by Pradit kumara and Ranjitha kumara (2010) were reported that medium containing 0/05 mg/l NAA cause increasing the root in Artemisia vulgaris. Result also showd MS medium with intraction of 2/0 mg/l NAA and 3/0 mg/l BA was effective on callus induction (more than 80 percent) that is because of CK effectin increasing cell division and auxin in retranscription differentiated cells to the meristematic cells.
