The rapid evolution of industry ۴.۰ and the emerging paradigm of industry ۵.۰ have transformed the architectural discipline, introducing technologies capable of bridging the gap between design intent and operational performance. Among these innovations, the Digital Twin-a dynamic, data-driven virtual replica of a physical asset-offers unparalleled opportunities for real-time monitoring, simulation, and adaptive decision-making. While Building Information Modeling (BIM) has been widely adopted, it remains a predominantly static design tool, lacking the bidirectional feedback and predictive capabilities required for resilient and sustainable architecture. This study proposes a novel framework for applying Digital Twin technology to adaptive and multis-species architecture, extending the concept beyond human-centered design to encompass biodiversity, ecological interaction, and environmental resilience. Using a mixed-method approach that combines conceptual modeling, IoT-based environmental sensing, and Al-powered simulation, the framework enables continuous optimization across the building lifecycle-from design and construction to operation and regeneration. A case scenario demonstrates how integrating real-time environmental and occupancy data can enhance thermal comfort, energy efficiency, and ecological compatibility. The findings suggest that Digital Twin systems can act as active agents of architectural adaptation, fostering both sustainability and multi-species coexistence. This research positions Digital Twin technology not only as a performance management tool but as a transformative paradigm for architecture in the age of sustainability.
