Investigating the Ability to Induce and Produce Hairy Roots by Different Strains of Rhizobium Rhizogenes Bacteria in Damask Rose (Rosa Damascene Mill.)

Damask rose (Rosa damascena Mill.) is one of the most important aromatic roses used in the production of perfumes, essential oils, and medicinal products. Enhancing the production of secondary metabolites is possible through the application of modern biotechnological techniques, such as culturing hairy roots. Hairy roots induced by Rhizobium rhizogenes are suitable tissues for secondary metabolite production due to their genetic stability and high growth rate under hormone-free conditions. In the present study, different strains of Rhizobium rhizogenes (ATCC۱۱۳۲۵, ATCC۱۵۸۳۴, A۴, A۷, A۱۳, and K۵۹۹) were used to induce hairy roots from complete seedlings and leaf explants of Digitalis purpurea. The explants were cultured in half-strength MS medium (½ MS). Hairy roots appeared ۲ to ۳ weeks after bacterial inoculation. The highest frequency of transgenic root induction (۶۰%) was observed in leaf explants inoculated with the A۴ strain. In complete seedling explants, no hairy roots were observed, except in those treated with the A۴ strain.
