Stimuli-responsive smart nanoparticles have emerged as a transformative platform for targeted cancer therapy by enabling controlled drug release in response to specific environmental cues. These nanocarriers are engineered to detect endogenous stimuli such as acidic pH, redox gradients, enzyme activity, and hypoxia within the tumor microenvironment (TME), as well as exogenous triggers like temperature, light, and magnetic fields, allowing precise and effective therapeutic delivery. Smart nanocarriers enhance tumor accumulation, reduce systemic toxicity, and improve therapeutic outcomes by responding to both single and multiple stimuli. Recent advances include polymeric, liposomal, and multifunctional nanoplatforms with optimized release kinetics and targeting efficiency. However, challenges such as clinical translation, large-scale manufacturing, safety, and regulatory hurdles remain significant. This review summarizes the design principles, operational mechanisms, and current progress in stimuli-responsive nanomedicine for cancer therapy, highlighting future directions toward clinical applications.
