Ferulic acid improves seizure severity through inflammatory factors and oxidative stress in a maximal electroconvulsive seizure model in mice

Ferulic acid (FA) is a phenolic compound with antioxidant, anti-inflammatory, and neuroprotective effects. This study evaluated the therapeutic potential of FA in the maximal electroshock-induced epilepsy (MES) model in mice by measuring total antioxidant capacity and interleukin-۱beta levels. In this study, ۶۰ male mice were randomly divided into ۶ groups. Pretreatment with FA (۲۰ and ۸۰ mg/kg/day) was started intraperitoneally four days before maximal electroshock (MES) seizure induction. Adverse effects of the drugs were assessed using the chimney test. Interleukin-۱β (IL-۱β) and total antioxidant capacity (TAC) levels were measured in the brains of mice following seizure induction. Ferulic acid administration at both doses of ۲۰ and ۸۰ mg/kg significantly reduced hindlimb extension (HLE) duration compared to the control group. In addition, ferulic acid at both doses increased TAC levels and decreased brain IL-۱β after seizure induction. The drug did not cause any adverse effects on motor coordination in mice. The results of this study indicate that pretreatment with FA can be effective in improving MES-induced seizures in mice by regulating brain TAC and IL-۱β levels.
