Molecular Study of Lys Gene for the Production of L-Lysine Amino Acid from Thermophilic Bacilli in Susceptible Cells for Use in Animal and Poultry Feed

Background and aim: Bacillus thermophilus is a type of bacillus that carries various genes, including the gene for the production of the essential amino acid of lysin (Lys). The aim of this research is the molecular study of the Lys gene for the production of L-lysine amino acid from thermophilic bacilli in susceptible cells for use in livestock and poultry feed. Materials and Methods: Bacillus strains were isolated and identified from a total of ۱۸ soil samples from the desert areas of Kerman sent to Pasargad research laboratory, and then Lys genes were extracted from these bacteria by PCR method. The amplified fragment was transformed and cloned by pTG۱۹ vector into E.coli uragami bacteria and cloning was confirmed using common methods and a phylogeny tree was drawn.  Results: PCR reaction for Lys genes was performed with the mentioned primers. Out of ۱۲ isolates of thermophilic bacillus found, ۳ isolates (۲۵%) carried Lys gene. The presence of the mentioned gene was verified by real time PCR test and PCR product sequencing confirmed the cloning of L-lysine in Escherichia coli Oragami. Conclusion: The appropriate level of Lys in the diet leads to optimal growth and health in poultry, in this study, the potential of thermophilic bacillus isolated from soil was evaluated as a rich source of secondary metabolites, and the native strain was used to produce acid. The essential amino acid L-lysine was investigated and the L-lysine enzyme was amplified through the ptg۱۹ vector and TA cloning technique. As a result of the present study, it seems that the native Bacillus thermophilus can be used due to its high potential in the production of L-lysine and also in line with Optimization and microbial synthesis of the aforementioned essential amino acid is beneficial for its use in various uses, especially in the food composition of livestock and poultry and to meet their nutritional needs.
