Purpose: Doxorubicin (DOX) is one of the most common chemotherapy drugs that is used in different types of cancer, but it causes fertility problems in cured patients. Combining doxorubicin with therapeutic antioxidants is one of the ways to reduce oxidative stress in other organs. Empagliflozin (EMPA) is a drug with antioxidant properties in the treatment of type ۲ diabetes mellitus. The aim was to investigate the effects of EMPA as an antioxidant on doxorubicin induced ovarian toxicity in NMRI mice. Method: ۲۴ female NMRI mice with ۶-۷ weeks' age were randomly divided into four groups, and during the ۲۸-day period, the DOX group received a ۱۰mg/kg i.p. single injection, the DOX+EMPA group, in addition to receiving a single dose of doxorubicin, during the ۲۸-day period received ۱۰mg/kg/day i.p empagliflozin, the EMPA group received ۱۰mg/kg/day i.p empagliflozin and the control group did not receive any drug. Ovaries were collected for histopathological analysis, and serum concentrations of sex hormones, malondialdehyde (MDA) and the ferric ion reducing antioxidant power (FRAP) were measured. Results: In DOX group, doxorubicin decreased factors such as the number of follicles, weight of body, total antioxidant capacity, sex hormones and increased blood sugar and lipid peroxidation. In the DOX+EMPA group, compared to the DOX group, empagliflozin, improved most of the mentioned factors. Conclusion: Our results showed that empagliflozin, by improving the antioxidant defense system can reduce stress oxidative and prevent the adverse effects of DOX in female NMRI mice.
