Sorbus pohuashanensis is a precious tree species with ornamental, medicinal, and edible properties, having deeply dormant seeds. Saline-alkaline stress is a major environmental factor affecting plant growth and development. The impact of saline-alkaline stress on embryo germination and its regulatory mechanisms remains unclear. Saline-alkaline tolerance in plants can be enhanced through genetic and non-genetic methods. Exogenous substances such as nitric oxide (NO), polyethylene glycol (PEG), and plant growth regulators like gibberellic acid (GA) and ethylene (ETH) are commonly used to improve saline-alkaline tolerance. This study investigates the effects of PEG and Ethephon (ETH donor) on the germination of S. pohuashanensis embryos under saline-alkaline stress. The study records the germination rate of embryos and analyzes hormone content, reactive oxygen species (ROS), antioxidant enzyme activity, and reactive nitrogen species (RNS) content. The findings provide insights into the mechanisms of ETH and PEG in mitigating salinity stress during S. pohuashanensis embryo germination, laying the groundwork for further research on exogenous regulatory substances to enhance saline-alkaline tolerance.
