Malling Merton۱۰۶ (MM۱۰۶) is known as one of the most valuable semi-dwarf apple rootstocks in the world. This study was evaluated micro propagation MM۱۰۶ rootstock through in vitro tissue culture technique. Sampling was carried out from lateral buds at four different times and locations. Evaluations of means about uncontaminated explants at index sampling show that there is best result in sampling (B) with ۸۰% success. Also to prevent the of production phenolic compounds in the establishment stage explants were treated with cold and darkness care, and then explants after five weeks were prepared to enter the shoot proliferation stage. Explants in the shoot proliferation stage were taken from shoot tips or single nodes, and then in this stage was used of Murashige and Skoog (MS) medium containing benzyl adenine (BA), indole-۳-butyric acid (IBA), gibberellic acid (GA۳), phloroglucinol (PG), agar and sorbitol. The first stage of rooting was performed for a week on MS medium with half normal concentrations of mineral salts, together sorbitol and PG, but without BA and GA۳. Also, two common auxins including indole-۳-acetic acid (IAA) and IBA were used separately or in combination. In the second stage of rooting was used of MS medium without auxins and PG for four weeks. Eventually, at the end of five weeks, explants were evaluated at indices of root length, root number, and rooting percentage. Comparison of means at index roots number shows that in treatments of T۴ = ۵/۵ and T۶ = ۵/۶, there are significant differences at the ۵% level. Also, a comparison of means at index root length shows that in all of the treatments, there are no significant differences at the ۵% level, but in treatment of T۱=۱/۳۵cm shows the highest root length. Finally, analysis of means at index rooting percentage shows that in treatments of T۳, T۴, and T۶ there is the best results with one hundred percent.
