Analysis of candidate genes and enzyme activities associated with defense responses to stem canker disease caused by Rhizoctonia solani Kühn in potato

Rhizoctonia solani Kühn is one of the most important fungal pathogens of potato (Solanum tuberosum L.). Resistant sources are the only effective method of control for managing the disease. In this study, ninety-two potato genotypes were screened against R. solani under field conditions of which, ۳ resistant and ۳ susceptible potato genotypes were subjected to bio-genetic assays using: PR-۵, Glub۲, pcht۲۸, PR-۱b, ERF۴, PAL۱ and LOX genes; and peroxidase, superoxide dismutase, polyphenol oxidase, catalase and phenylalanine ammonia-lyase enzymes analysis. The resistance and susceptibility of the ۶ re-inoculated resistant and susceptible potato genotypes were reconfirmed under greenhouse conditions. The results reveled that, biomass scales in susceptible potato genotypes decreased more compared to resistant genotypes. Changes in expression levels of PR-۵, Glub۲, pcht۲۸, PR-۱b, ERF۴, PAL۱ and LOX genes indicated increases in defense and activities of the antioxidant enzymes; peroxidase, superoxide dismutase, polyphenol oxidase, catalase and phenylalanine ammonia-lyase. There was an increase in gene expression and enzyme activities in inoculated resistant potato genotypes compared to susceptible and control genotypes. Given the changes in biomass parameters, gene expression level and enzyme production rate, further steps towards identification and production of potato cultivars resistant to R. solani fungi will be possible.
