Conventional cancer treatment methods such as chemotherapy and radiotherapy often induce side effects on normal cells and may elicit resistance in cancerous ones. Furthermore, prostate cancer cells can develop resistance to DTX treatments. In the recent years, researchers increasingly tend to utilize radiosensitizers such as silver nanoparticles (AgNPs) alongside with low dose X ray irradiation to mitigate radiotherapy side effects. This study investigates the anti tumor effects of a combination therapy involving DTX loaded onto alginate coated AgNPs along with radiotherapy on prostate cancer cell line (LNCaP).AgNPs were synthesized using a green method with Caucasian whortleberry fruit extract as a reducing agent. These nanoparticles were then coated with alginate to facilitate DTX loading. The physiochemical properties of Ag@AlgDTX NPs were characterized using FT IR, DLS, FE SEM, TEM, hemolysis, and release tests. Cellular uptake of NPs was assessed via flow cytometry test. Finally, anti tumor investigations were carried out through MTT, apoptosis and scratch assays on LNCaP cells.The physiochemical assays confirmed the size, shape, and successful synthesis. Drug release assay was timedependent with the peak release at ۲۴h. Ag@Alg DTX NPs were efficiently internalized by LNCaP cells. Moreover, the radiosensitizing effect of AgNPs notably enhanced the efficacy of X ray irradiation. The combination therapy significantly inhibited cell growth and induced apoptosis.Results demonstrated that the combination therapy using Ag@Alg DTX NPs and radiotherapy holds promise for apoptosis induction and cell growth inhibition in prostate cancer cells.
