Introduction: Studies have shown that wormwood plant containsmore than ۶۰۰ secondary metabolites. Artemisinin is asesquiterpene endoperoxide molecule. Because of the being,۱,۲,۴ thyroxane bridge is one of the most active secondarymetabolites of wormwood plant. Due to the toxic free radicalsproduced by the endoperoxide part,it plays a significant role inthe specific death of cancer cells and inhibition of tumor angiogenesis.In this study, the wormwood extract and artemisinin asits active ingredient was used in the diet of mice. Then the geneexpression profile related to proliferation and invasion of coloncancer cells was investigated. Studies have shown that the highexpression of lncRnaNEAT۱ is significantly related to the initiationand progression of colorectal cancer.Materials and Methods: Three groups of mice under standarddiet (N=۵, Control Group), wormwood extract (N=۵, TreatmentGroup) and artemisinin (N=۵, Treatment Group) were treatedfor ۲۱ days. Then intestinal tissue was separated and completeRNA extraction was performed. After cDNA synthesis, changesin the gene expression was performed using specific primeragainst lncRNANEAT۱ gene were investigated by Real timeRT PCR method. Eventually relative expression changes of thedesired gene were calculated based on the ۲-ΔΔCT method.Results: Our results showed the expression of lncRNANEAT-۱gene in two treatment groups under wormwood extract and artemisininactive ingredient decreased to one third and a half, respectively,compared to the control group(Sig.= ۰.۰۰, P≤۰.۰۵).Conclusion: The use of plant compounds in the diet can playa significant role in the changes in the expression of the genesof the intestinal wall.Our results showed that the active ingredientsin this extract, which is primarily artemisinin, which actsas an inducer in the changes in the expression of genes for thebenefit of preventing Colorectal cancer is useful.
