Quercetin nanoliposomes protect acute low-dose diazinon neurotoxicity and behavioral impairments by affecting GABA receptors in the male rats

Diazinon (DZN) is an organophosphate insecticide that is widely used. We studied the effect of quercetin pegilated nanoliposomes (QPEGNL) on acute low level DZN-induced neurotoxicity and behavioral disorders by affecting brain gamma-aminobutyric acid (GABA) receptors for the short term in male rats.The pegilated quercetin nanoliposomes were prepared by the thin film evaporation. Animals were divided into five random groups included normal and vehicle control groups. Three groups were received DZN (۱۰ mg/kg, i.p.), two DZN- received groups were treated with quercetin QPEGNL ۲۰ mg/kg and quercetin ۲۰ mg/kg for ۲۴ hours. Behavioral experiments on the open field (OP) were investigated, and the total distance moved and mean velocity parameters were considered as indices for motor activity measurement. Animals were decapitated, brains removed, and frozen at -۸۰. The brain GABAA receptors expression was determined by qRT-PCR for anxiety-like behavior estimation.In results, total distance moved and mean velocity parameters significantly decreased in the DZN group than the control one (p < ۰.۰۵), and in the QPEGNL+DZN group increased than the DZN group significantly (P < ۰.۰۵). The expression of the GABAA receptors in the DZN group was reduced to the control one differently (P < ۰.۰۵) and in the DZN+QPEGNL group improved to the DZN group (P > ۰.۰۵).In this study, we concluded that low dose of DZN affected the brain GABAergic system and reduced acutely expression of GABA receptors which led to anxiety-like behaviors such as decreased motor activity or exploratory behavior although it was improved by QPEGNL.
