Background: Utilization of antioxidants in semen diluent, protectscells from oxidative damages. Supplementation of coolingmedium with mitochondria targeted antioxidants could bea beneficial way to improve semen quality during chilling process.The current s tudy was aimed to assess the influence ofusing Mito-TEMPO as a mitochondrial targeted antioxidant onquality parameters of buck semen during cooling processes.Materials and Methods: In this experiment, ۵ Saanen buckswere used and semen samples were collected twice a week.After semen collection and primary evaluation, diluted semensamples were added to extenders containing ۰, ۱, ۱۰, ۱۰۰, ۱۰۰۰μM Mito-TEMPO. The prepared samples were s tored at ۵ ◦Cup to ۴۸ hours. Chilled sperm viability, mitochondrial membranepotential and membrane functionality were assessed during۰, ۲۴ and ۴۸ hours.Results: In results, at time ۰, no difference was observed amonggroups but at ۲۴ and ۴۸ h s torage, higher (P ≤ ۰.۰۵) sperm mitochondrialactivity, membrane functionality and viability wereobserved in ۱۰ and ۱۰۰ μM Mito-TEMPO groups than the othergroups.Conclusion: Therefore, supplementation of cooling mediumwith Mito-TEMPO (۱۰ and ۱۰۰ μM) could be an efficientmethod to improve the quality of buck's semen during s torageperiod.
