Background: The action of vitamin D (VitD) on male reproductiveperformance has been controversial mainly because itis difficult to dis tinguish between what is mediated by calcium/phosphorus homeos tasis and what is due to genomic and nongenomiceffects. In this regard, rodent models in which VitDdeficiency (VDD) was artificially induced without creating animbalance in calcium and phosphorus homeos tasis showed thatsperm s tructure and function were affected and that the intensityof the alteration followed the severity and duration of theVDD.to show the genomic and non-genomic effect of VDD onspermatogenesis.Materials and Methods: Tes ticular extracts from mouse modelsof VDD with different degrees of severity, obtained eitherthrough the use of a vitamin D-deficient diet or paricalcitol injections,were analyzed by QRT-PCR to assess the s teady-s tateRNA levels of the major s teroidogenic players, including CYP۲۴A,CYP۱۹A۱, ۱۷ßHSDH۱, ۳ßHSDV۱, as well as es tradiol,tes tos terone, LH and FSH levels. In addition, tes ticular his tomorphometricand sperm s tructural and functional parametersanalyses were carried out.Results: we show that the accumulation of transcripts of eachmonitored s teroidogenic actor decreases linearly with the severityof the VDD. We also show that already in situations oflow and medium VDD, the es tradiol and tes tos terone content oftes ticular extracts is s trongly decreased.Conclusion: VDD exerts potent effects on tes ticular s teroidogenicsupport of spermatogenesis, independently of its wellknowncalcitrophic action. Therefore, VDD-mediated reductionof s teroidogenesis is clearly an aspect of the loss of tes ticularspermatogenetic efficiency in vitamin D deficiency, a very commonsituation worldwide.
