Isolation and identification of Mycobacterium from poultryand personnel of Bird’s Garden of Alborz province

چکیده مقاله:
Background and Aim : Mycobacterium genus causes many important diseases in humans,livestock and poultry. Non tuberculosis mycobacteria (NTM) are considered as atypicalmycobacteria, some of which cause important diseases. Mycobacterium avium complex (MAC) akind of NTM, causing casualties in immunosuppressed patients with Acquired immunodeficiencysyndrome (AIDS). According to the report of the World Health Organization, the second infectiouscause of death after Covid-۱۹ virus disease is tuberculosis, and the second cause of death in AIDSpatients is mycobacteria, especially the Mycobacterium avium complex (MAC) group. Therefore,investigating the presence of these mycobacteria in the population of birds and personnel anddetermining their exact identity helps public health, especially in AIDS patients.Methods : Near ۱۰۰ samples from personnel’s sputum and poultry feces of Alborz bird’s gardenwere collected, decontaminated by HPC and NaOH method, then cultured on mycobacterialmedium like Lowenstein-Jensen. The Mycobacterial genomic DNA was extracted by Phenol-Chloroform extraction method. By using ۱۶srRNA PCR method, Mycobacterium genius detectedand three kinds of its species identified by their specific primers like IS۹۰۰-۹۰۱,IS۶۱۱۰ and IS۹۰-۹۱.Results : In ۴۵ bird’s samples, ۱۵ samples were detected as Mycobacteria. Among those ۱۵samples, TB complex and NTMs such as Mycobacterium avium complex were identified.Conclusion : The result in this study indicated that different kinds of important mycobacteria wereisolated from specimens. Hygiene measurements should be taken to prevent these bacterialtransmission to people, especially people with immune deficiencies.
