Sestrin۲ is known as the markers involved in progression of aging. Exercise training, on the other hand, is known to be one of the most important modulators of aging, however, the effects of high intensity interval training exercise (HIIT) on Sestrin۲ are less well known. Therefore, the aim of this study was to compare the effect of high intensity interval and continuous training on skeletal muscle sestrin۲ in elderly rats.Thirty old male Wistar rats (۱۸ months) were randomly divided into control (C) (n=۱۰), high intensity interval training (HIIT) (n=۱۰), and continuous training (MICT). After estimating the maximum speed, rats in HIIT group participated in ۸ weeks of high-intensity interval training of running on the treadmill (۵-۸ sets of running for ۲ minutes at intensity of ۸۰-۱۰۰ VO۲Max with rest intervals of running for ۲ minutes at intensity of ۵۰ VO۲Max) five sessions per week and rats in MICT groups participated in continuous training protocol included (۸ weeks/ ۳ sessions per week with ۶۵ to ۷۰% VO۲max for ۴۰ min) running on a treadmill. Skeletal muscle tissue samples were extracted ۴۸ hours after the last training session to measure protein levels of factors assessed. Independent t-test was used to analyze the data and P<۰.۰۵ was considered significant.The results showed that HIIT resulted in a significant increase in sestrin۲ levels compared to the control group (P<۰.۰۵). However, MICT protocol had no significant effect on the protein values of sestrin۲ (P <۰.۰۵). Also, there was no significant difference between the two training groups (P <۰.۰۵).It seems that HIIT can play more effective role in increasing the protein levels of sestrin۲, which plays a central role in progression of aging.
