چکیده مقاله:

Cisplatin is one of the most potent chemotherapeutic drugs widely used to treat cancers.However, its clinical use is limited due to irreversible ototoxicity as well as the incidence ofrenal impairment. Therefore, one of the strategies to overcome the above problems is to designnew anti-tumor agents associated with cisplatin to change the nature of the metal ion. Among thenon-platinum metal complexes studied for cancer treatment, palladium(II) derivatives are havereceived a lot of attention. In this study, the cytotoxicity of bisdiphenylphosphine palladium(II)complexes (C۱-C۵), which were synthesized previously, was investigated against ovarian cancer(SKOV۳), lung cancer (A۵۴۹), and breast cancer (MCF-۷) cell lines. Based on MTT assayresults, the most active compound, C۳, showed promising cytotoxic activity with IC۵۰ values of۴۲.۷۱, ۳۹.۱۷, and ۱۰.۵۸ μM on lung, ovarian, and breast cancer, respectively. Furthermore, theinteraction of compounds with DNA structure (PDB code: ۱BNA) was also investigated bymolecular docking. Docking results indicated that C۳ is located in the minor groove of DNA andvia its nitro group interacts with C۱ base through hydrogen bond. The phenyl ring and G۲ basepair take apart in π-π interaction. Moreover, Cl atoms make polar interactions with G۲ base pair.Overall, these findings demonstrated that these complexes could be used as potential candidatesfor drug development in the future.
