The effect of glycyrrhizin acid on Bax and Bcl۲ expression in hepatotoxicity induced by titanium dioxide nanoparticles in Rats

Aim: This research studied the effects of GA on apoptosis induced with NTiO۲ in the liver of rats.Background: It is widely accepted that the contaminations resulting from nanoparticles (NPs) is an emerging dangerous issue. Metal oxide nanoparticles have high environmental stability and cause toxicity in the food chain. Thus, the present study investigated the anti-apoptotic effects of glycyrrhizic acid (GA) on the hepatotoxicity generated by titanium dioxide (NTiO۲) NPs in the liver tissue. Methods: Thirty-two male Wistar rats were randomly divided into four groups. Control group, received saline by gavage for ۲۱ days. NTiO۲-treated rats were given ۳۰۰ mg / kg of NTiO۲ solution via gavage for ۱۴ days; GA-treated were administered ۱۰۰ mg/kg GA for ۲۱ days; protection group was pre-treated with GA before NTiO۲ administration for ۷ days. Then, apoptotic index was evaluated through immunolocalization of Bax and Bcl-۲ and TUNEL assay.Results: we found that HSCORE of Bax expression and apoptotic index experienced a significant increase whit NTiO۲ (P <۰.۰۰۱) while Bcl-۲ expression significantly diminished in NTiO۲-treated rats (P <۰.۰۰۱). The results revealed that the increase Bax expression and apoptotic index was reversed by GA and enhanced the activities of Bcl۲.Conclusion: The results revealed that GA effectively attenuated apoptosis against NTiO۲ in rats.
