چکیده مقاله:

Immobilization of bio-molecules on substrate is the most important in bio-conjugation and medical industry. In most time, the structure of bio-molecules was changed or their functionality was loose, in immobilization step. Also, we can optimize the immobilization factors for achievement of high efficiency. In this study, we immobilized T۴ bacteriophage on gold electrode by streptavidin/biotin linker for E coli bacteria detection. This immobilization has two steps: a) immobilization of streptavidin on gold electrode and b) attachment of biotin on bacteriophage surface. In first step, the streptavidin by EDAC/NHSon gold electrode then, the biotin modified bacteriophage conjugated by streptavidin. The results are studied by SEM. Then, the bacteriophage immobilized on gold electrode biosensor was patterned as a two electrodes configuration. Consequently, the E. coli trapping by oriented bacteriophage biosensor was reported by surface plasmon resonance (SPR) method. We successfully developed and implemented an E. coli biosensor based on bacteriophageimmobilization on Au electrodes. This biosensor is cost effectiveness, rapid, renewable and selective for detection of E. coli bacteria. Also, this method can be extension to detection of other biological agents.
