بررسی جداسازی و تخلیص پروتیین ایمنودومیننت ‮‭34‬ کیلودالتونی مایکوباکتریوم آویوم زیر گونه پاراتوبر کلوزیس

Johne's disease or Paratuberculosis is a chronic granulomatous inflammation of the small intestine of cattle and other ruminants, caused by infection with Mycobacterium avium subspecies paratuberculosis (M. paratuberculosis ) The disease can be found in ruminant herds worldwide, causing substantial economic losses at farm level due to premature culling and production losses M. a. ptb is a acid-fast rod which forms rough nonpigmented colonies when grown on artificial media, such as Herrolds egg yolk, Watson- Reid or Lowenstein-Jensen medium. Proteins secreted by Mycobacterium species have been suggested as major immune targets in the early phase of infection. In this study We here describe the identification and characterization of novel Mycobacterium avium subsp. Paratuberculosis lll-V strain antigens by Preparation of culture filtrate and soluble cellular extracts. we have used SDS-PAGE technique to index the antigenic contents of culture filtrate and bacterial extracts of bacteria. The largest number of bands 17weighed 19-100 kDa, In this study we could to detect of desired protein(34 KDa) by SDS Page and cut desired rang of 34KDa. Immunology results showed that we can use of this Protein to develop of Kit to detect of bovine paratuberculosis.
