تکثیر غیرجنسی شاه بلوط به روش کشت بافت

In order to investigate asexual propagation of Castanea sativa by tissue culture techniques, apical and axillary buds taken in different seasons from selected mature trees at Shafarood forests in Gillan were used as explants. After different sterilizing treatments, they were cultured on DKW, QL and MSN/2 mediums. Late autumn was the best season for explanation and using 0/01% mercuric chloride for 7 minutes proved to be the most effective sterilizing treatment. DKW medium containing BAP 0.5 mgl-1 + IBA 0.1 mgl-1 was the best treatment for establishment. In shoot multiplication DKW medium containing BAP 0.5 mgl-1 + IBA 0.01 mgl-1 achieved the best rate of proliferation and elongation. Rooting of shoots by in vitro has been done in two methods: 1- 2 cm long shoots in MS medium (و nitrate) containing 3 mgl-1 IBA for 12 days cutured and then transferred to an auxin-free medium after 4 weeks 83% shoots rooted. 2- The shoots were treated as cuttings by dipping the basal ends in 50 mgl-1 IBA solution for 24 hours after which they were placed in an auxin-free. GD medium (1/3 macro element) plus AC. After 4 weeks 100% of shoots were rooted. Rooted plants after acclimation transferred to soil under greenhouse condition.
