EVALUATION OF HTR2A RS6313 POLYMORPHISM AND HEROIN DEPENDENCE SUSCEPTIBILITY AMONG POPULATION FROM NORTHWEST OF IRAN

Background and Aim : Heroin dependence is a chronic relapsing disorder that caused by a combination of genetic, epigenetic, environmental factors and it is a big public health problem all over the world. Previous studies have shown that the genetic risk of addiction is approximately 40-60%. Variation in serotonin (5-HT) transmission in the central nervous system has been implicated in alcohol and drug dependence. Recently studies show that the serotonin type 2A gene (HTR2A) T102C/rs6313 polymorphism play a significant role in alcohol dependence. Here we evaluate the association of HTR2A rs6313 polymorphism and heroin dependence in the northwestern population of Iran.Methods : In this case-control study, the case group consisted of 100 heroin addicts and the control subjects consisted of 102 men with no history of substance and analgesic drugs use and no familial history of drug abuse. DNA extraction was done from peripheral blood samples by using the salting-out method. Genotypes were determined for HTR2A rs6313 in case and control groups by PCR-RFLP technique. Msp1 restriction enzyme employed for genotype discrimination.Results : The Frequencies of the genotypes CC, CT and TT was 23%, 50% and 27% in patient group and 32.35%, 44.12% and 23.53% in control group respectively. The statistical analysis showed no significant association between HTR2A rs6313 polymorphism and susceptibility to heroin dependence in population of northwest of Iran (p> 0.05). Conclusion : Our results propose that HTR2A rs6313 may not contribute to the susceptibility of heroin dependence in the Northwest of Iran male population.
