Novel 99mTc-Dendrimer-Glycoconjugate as Heart Diagnostic Agent

Background: Today, researches focus on providing novel, safe and cost-effective molecular imaging radiopharmaceuticals. To date,18FDG is frequently used for myocardial viability scan. But the short half-life of 18F (109.8 min), and high cost of 18FDG has limited molecular studies. In the present experiment, 99mTc-anionic citric acid based-PEG dendrimer-second generation-glucosamine (99mTc-nano-conjugate) was designed for myocardial viability scan.Method: Dendrimer-G2 was synthesized and conjugation of glucose and dendrimer was prepared thereafter. Toxicity on normal cells was assessed by XTT assay. radiochemical purity (RCP) was performed by chromatography. Imaging scan was done with SPECT/CT. Three rabbit and mouse was chosen.Results: The synthesis of the dendrimer and conjugation were confirmed by FT-IR, 1HNMR, LC-MS, DLS. XTT assay showed no in-vitro cytotoxicity. Based on data yield by the RCP, the anionic dendrimer has a very high potential to complex formation with 99mTc and also is very stable in the human serum in different times. In-vivo study including SPECT and biodistribution were done to confirm the potential imaging of the 99mTc-nano-onjugate.Conclusion: Generally, suitable features of the dendrimer including biocompatibility, biodegradability, low toxicity, solubility, low cost, stability and chelator-free radiolabeling of dendrimer with 99mTc, show that it is a promising agent to improve nano-radiopharmaceutical.
