Preparation and in-vitro evaluation of Pomegranate extract- nanogel for topical treatment of oral ulcers

Introduction and background: Pomegranate (Punica granatum), has been extensively used in the folk medicine of many cultures, possess antioxidant, anti-inflammatory, and anti-bacterial properties. Nanogels (NGs) are currently under extensive investigation due to their unique properties, such as small particle size, high encapsulation efficiency and protection of active agents from degradation, which make them ideal candidates as drug delivery systems.Methods: pomegranate nanoparticles were prepared by anti-solvent precipitation method. Ethanol and water were selected as solvent and anti-solvent, respectively. PVA was used as hydrophilic polymer and chitosan was used as hydrophobic polymer. Tween-80 was selected as a surfactant to inhibit the particle growth and control the particle morphology.Results: In this research, pomegranate extract-encapsulated core-shell type nanoparticles were prepared. Obtained results of Zeta potential and DLS analysis revealed that the particle size of pomegranate nanoparticles was around 200 nm and the morphology was spherical shapes. Pomegranate nanogel was active against S. sanguis, S. mutans and S. mitis by controlling their adhesion potential to the surface in the oral cavity.Discussion and Conclusion: Pomegranate nanogel possesses anti-bacterial and anti-fungal therapeutic effects on oral pathogens. These treatments are aimed at prevention of secondary infection, reduction of pain and inflammation, and promote healing of ulcers. Pomegranate nanogel can be used in treatment of varied range of oral ulcers such as, aphthous stomatitis, lesion, and periodontitis
