Background and Objective: Gestational diabetes mellitus is defined as impaired carbohydrate and lipid metabolism. During pregnancy and is characterized by progressive hyperglycemia and insulin resistance and compensatory hyperinsulinemia. Several studies have reported metabolic and genetic disorders in women with gestational diabetes mellitus (GDM). This study was carried out to evaluate the effects of a novel probiotic supplementation diabetes mellitus. Methods and Materials: In a randomized, double-blind, placebo controlled clinical trial, 48 patients with GDM. particpants were randomly divivided into two groups, each group receiving two of the following daily supplement probotic (n=24) or placebo (n=24) for six weeks. Sampling were done before and after Intervention. After RNA Extraction and cDNA synthesis, Changes of genes expression were measured by real time PCR. Results: Our results showed that, Probiotic inrtake for 6 week, upregulated peroxisome proliferator- activated receptor gamma (p=0.01), and downregulated gene expression of tumor necerosis factor alpha (p=0.03) in peripheral blood mononuclear cells of subject with GDM. There were no statistically significant changes following supplementation with Probiotic on genes expression of Interleukin 1( IL-1) , Interleukin 8 (IL-8) , Oxidized low-density Lipoprotein receptor(LDL-ox), Lipoprotein a ( Lp-a ) and Glucose transferase1 (GLUT-1) in PBMCs of patients with Gestational diabetes. Conclusion: Overall, probiotic supplementation for 6 weeks to patients with GDM had beneficial effects on genes expression. PPAR-y and TNF-a
