چکیده مقاله:

Introduction and Objectives: Mycobterium tuberculosis has infected more than two billion individuals worldwide, out of whom 5-10% have clinically active diseases and 90-95% remain in the dormant form with viable bacteria in the macrophages as their reservoir for extended periods of time. The dosRregolone consists of approximately 50 geneswhich are essential for survival in the latent period . With this regard, there is no specific treatment to target people with this condition, so the activation of latent MTBwill always remains a threat to them. Materials and methods:1-Microscopicditections2-Culture of Mycobacterium3- DNA extraction using CTAB method 4-DNA electrophoresis 5- Detection of Mycobacterium tuberculosis Complex Species Using PCR-IS6110 6- Determination of drug sensitivity by multiplex PCR for isolation of MDR-XDR-TDR-S mycobacteria. 7-Creating a dormancy state for genes under zero oxygen conditions8- Primer design and PCR 9- Digest the PCR products of all genes using their specific enzymes ( devR: Acil Enzyme &devSEaeI Enzyme ) by PCR-RFLP 10- Evaluation of results on polyacrylamide gel Results:Electrophoresis PCR Product revealed two specific bands of 418bp for devR and 767 bp for devS genes .Digestionof PCR Product by Aci enzyme for devR gene (20-50-100), and EaeI enzyme for devS gene (20-40-110-250-350-500-650), respectively were observed. Conclusion :Since the expression of dosRregolon increases in the latency stage, and T-cells show specific responses to some of the antigens which are specific to the regolon, it can be considered as a candidate for the production of a vaccine and drug.
