Evaluation of Immune Response of multi-epitope Recombinant Protein of Staphylococcus aureus as vaccine Candidate in Comparison with Natural Proteins of Hla, MntC and SACOL0723 of Staphylococcus aureus Standard Strain

Background and Objectives: Staphylococcus aureus infections are one of the most common causes of surgical drainage, bacteremia, and hospital-acquired infections. The emergence of antibiotic resistance has increased mortality and costs of treatment. The design of a new vaccine against S. aureus would have a great beneficial impact on public health. We aimed to evaluate the immunity of Hla-MntC-SACOL0723 multi-epitope protein of S. aureus in comparison with natural proteins of Hla, MntC and sacol0723.Materials and Methods: After recombinant, Hla, MntC and SACOL0723 proteins expression, proteins were purified by Ni-NTA column. The groups of mice were immunized with Hla, MntC and SACOL0723 proteins, designed epitope vaccine candidate and PBS. The functional activities of the proteins were evaluated by ELISA and opsonic killing activities, and also assay Th1, Th2 and Th17cytokines. Results: The study results indicated that our designed epitope vaccine could induce robust protective immunity responses. The fusion protein was able to elicit a mixture of Th1, Th2, and Th17 immune responses more towards Th1 and Th17 that are crucial for eradicating S. aureus infection. Conclusion: In conclusion, fusion protein admixed with alum adjuvant could be considered as potential vaccine candidates for protection against S. aureus infection.
