Interleukin-10 as a genetic predisposition factor for cervical cancer in patients with human papillomavirus

Introduction: Cervical cancer is the fourth most frequent female malignancy worldwide and the third leading cause of cancer associated death among women in less developed countries. The genetic factors had played an important role and one of the etiological factors for cervical cancer. The present study investigated the association between Interleukin-10 (IL-10) rs1800896 gene polymorphism and cervical cancer and aimed to detect the correlation between serum levels of IFN-γ, TGF-β, TNF-α and cervical cancer. Materials and Methods: in this case-control study. 146 patients with cervical cancer and 169 healthy controls who referred to Ghaem Hospital, Mashhad University of Medical Sciences, Mashhad, Iran were enrolled to study. The IFN-γ, TGF-β and TNF-α serum levels were measured by enzyme-linked immunosorbent assay (ELISA). Taq-man®‐probes‐based assay was used to detect genotype analysis of IL-10 polymorphism (rs1800896). Results: A significant association was found between IL-10 rs1800896 gene polymorphism and cervical cancer (P<0.001). The results of statistical analysis showed that the AA genotype could significantly increase the risk of Cervical cancer (P<0.001). Regardless of the genotypes, the serum IFN-γ, TGF-β and TNF-α levels were significantly higher in cervical cancer patients than in the controls (P<0.001). Additionally, a significant association was observed between IL-10 rs1800896 gene polymorphism and the serum level of IFN-γ, TGF-β and TNF-α (P<0.001). Conclusions: According to this study, IL-10 in genetic levels it might be a prediction factor for the disease. Furthermore, the serum level of TNF-α, TGF-β and IF-γ cytokines have directly correlated with cervical cancer.
