Study of frequency of G/A-308 polymorphisms in TNF gene among the infertile men with different stage of impaired spermatogenesis

Background: The tumor necrosis factor alpha (TNF-α) gene codes a cell signaling protein (cytokine) involved in systemic inflammation and is one of the cytokines that make up the acute phase reaction. The association of this gene polymorphisms on male fertility status is controversial. We evaluated the effect of G/A-308 polymorphism of TNF-α gene on semen stress oxidative and apoptosis parameters, sperm DNA damage and sperm chromosome aneuploidy rate among the Iranian infertile men. Methods: We used 210 men with infertility (Asthenospermia, oligospermia, oligo-asthenospermia) and 120 control participants. Results: We observed significant high frequency of A allele of this polymorphism among oligo-asthenospermic men)P<0. 01(. In semen samples of men with AA genotype, there was significantly high level of Malondyaldehyde, caspase-9 (P< 0.0001), and also we observed high frequency sperm DNA damage rate (P< 0.0001) and XY chromosomes disomy (P<0.0001) in sperm samples of men with AA genotype of TNF-α polymorphism. There was a significant correlation between sex chromosomes aneuploidy and sperm DNA damage rate in sperms of men with TNF-α polymorphism (R = 0.72, P, 0.048). Conclusion: Our investigation showed, G/A-308 polymorphism of TNF-α gene effect on stress oxidative pathway and causes sperm DNA damage and chromosomal aneuploidy and through this path can effect on fertility.
