Comparative analysis of four DNA extraction protocols for fresh and dried specimens of five medicinal Lamiaceae plants

Isolation of pure DNA from some medicinal plants is difficult due to high amounts of secondary metabolites produced by their cells. On the other hand, the genomic studies necessitate successful extraction of high-quality DNA [1, 2]. Hence, the present study aimed at comparing four methodologies of DNA extraction including Murry and Thompson (1980) [3], Dellaporta et al. (1983) [4], Doyle and Doyle (1990) [5] and Ziegenhagen et al. (1993) [6] from five genera of Lamiaceae. Both fresh and dried leaves of Rosmarinus officinalis, Lavandula officinalis, Salvia hydrangea, Dracocephalum kotschyi and Ziziphora tenuior were used for the procedures. After quantitative and qualitative analysis, the DNA preparations were tested by PCR amplification using two inter-simple sequence repeats (ISSR) primers. All of the procedures were effective for DNA extraction from all of the genera examined. Results showed that Murray and Thompson method was more efficient in terms of DNA quality and quantity (1.80 and 570.57 ng/μl DNA), respectively, compared with other protocols. We also observed that the quality and yield of extracted DNA from fresh leaves were relatively higher than those of dried leaves. In addition, banding pattern obtained via 1% agarose gel and PCR showed the same results. According to the data, Murray and Thompson method is recommended for DNA extraction from fresh and dried leaves of the five medicinal plants examined.
