Evaluation the effects of administration various doses of indomethacin on expression of inducible nitric oxide synthase in breast tumor

چکیده مقاله:
Introduction: Cancer is one of the major threats for human health throughout the world. It is currently the second cause of death. Breast cancer is the most common cancer among women in all over the world. Also breast cancer is an important public health problem. Indomethacin is a nonsteroidal anti-inflammatory drug that inhibits the production of prostaglandin. Due to the lack of study on the effect of indomethacin on breast cancer cells, in this study, the effect of various doses of indomethacin on the iNOS gene expression in breast tumors were investigated. Material and methods: Four groups of mice were considered. First group was healthy control group, second group was tumor control, the third and the fourth groups were under treated by various doses of indomethacin. Breast tumor cells were inoculated into mice after culture and proliferation. The rate of tumor growth was evaluated in tumor control group and the groups were under treated by various doses of indomethacin for two months with Real Time RT-PCR technique. Findings: The results showed that with the growth of breast cancer cells, iNOS expression in tumor tissue was increased. It could occur by different mechanisms such as immune response suppression in tumor environment in breast tumor growth have role. There is not any significant differences between two groups of study (P> 0.05). Results and discussion: In this evaluation we observed, administration high dose of indomethacin was toxic and had lethal effects on mice. 100mg and 25mg of indomethacin administration had no significant effect on breast tumor growth.
