Evaluate the Possibility of Induction of Neurogenic Differentiation of Mesenchymal Stem Cells Using Curcumin

Background and Aim: Neurodegenerative diseases are defined as diseasesthat cause impaired nervous system function and neurons death. Today,Scientists are seeking to use more effective, less costly and less harmfulsubstances for the treatment of neurodegenerative diseases. The purposeof this study was to examine neurogenic induction effects of curcumin on the bone marrow –mesenchymal stem cells (BM-MSCs).Methods: The BM-MSCs were extracted from rats. MSCs were culturedin a 96-parts plate to determine the curcumin toxicity. For evaluation ofneuron differentiation, at the first, BM-MSCs were cultured in 24 wellplates with DMEM medium containing 10% FBS. In the second day,pre-induction medium, including DMEM, BME, and FBS, was addedinto the cells. In the third day, plates were divided into two groups: theexperimental group and positive control group. The positive controlgroup was induced by two induction mediums separately: inductionmedium containing BME or BHA and DMSO. The experimental groupwas also induced by two inductive groups: curcumin induction groupand curcumin induction group with BHA and DMSO. After cellsinduction, every hour, each plate was photographed for six hours. Inthe final stage, after removing the medium from plates, both positiveand experimental control groups were placed in the DMEM mediumcontaining N2-supplement, butyl hydroxyanisole, potassium chloride,valproic acid, and forskolin.Results: The results of MTT showed that curcumin 1 to 10 μM did nothave any significant toxicity effects on BM-MSCs. The BM-MSCs weresuccessfully induced in two positive control groups that were treatedby BHA and BME, although the differentiation rate was higher by BHAinduction medium and the toxicity of this substance was lower than BME.Curcumin remarkably was able to induce neurogenic differentiation ofBM-MSCs, although this effect was noticeable increase when the cellswere treated by the combination of curcumin and BHA.Conclusion: Curcumin supposed to be a good drug for neurodegenerativediseases based on the induction of neurogenic differentiation
