Evaluating the Expression of Bone Gamma-Carboxyglutamate Protein in Differentiated Cells Subjected to Iron Nanoparticles

Background and Aim: Iron nanoparticles (NPs) in medical fields has beenutilized broadly in magnetic resonance imaging. It is crucial to formulateNPs containing appropriate characteristics for therapeutic approaches.The broad application of Mesenchymal stem cells (MSCs) in regenerativemedicine attracted the attention of scientists. The expression of thebone gamma-carboxyglutamate protein (OCN) as one of the markergenes of osteoblasts can be considered in the accuracy of inductionof the osteogenic differentiation. In the current study, we assessed theadverse effect of Zn-Mn-iron NPs on the expression of OCN gene in theosteocytes.Methods: In the current study, we exposed umbilical cord MSCs toosteogenic differentiation media. At day 21 the morphology of cells hasbeen altered to osteoblasts. The changes of OCN gene expression at themRNA level was evaluated by SYBR Green and real-time quantitativePCR.Results: The expression of OCN gene of treated cells with Zn-Mn FerriteNPs showed no significant changes in comparison with untreated cells.Conclusion: The results of our study showed that ferrite NPs do not haveany adverse effects on the expression of OCN gene at the mRNA level. Inorder to utilize ferrite NPs in therapeutic aspects, yet more investigationsare required to ensure the safe function of this approach.
