HISTOLOGIC, METABOLIC AND INFLAMATORY DERANGMENT IN THE LIVER OF HIGH-FAT DIET INDUCED OBESE RATS BEFORE AND AFTER VITAMIN D ADMINISTRATION

Background and Aim: Obesity is associated with numerous metabolic and inflammatory disorders.The current study was aimed to evaluated the effect of high_fat diet obesity on hepatic inflammatory, metabolic and histologic manifestations and also to investigate the effect of vitamin D administiration on the markers of inflammation and metabolic damages of liver in high_diet induced obese rats.Methods: In the beginning of the study, 40 rats were divided into two groups: control diet and high fat diet (HFD) for 16 week;then each group subdivided into two groups including normal diet(ND), ND+vitamin D , HFD, HFD+vitamin D. Vitamin D supplementation was done for 5 weeks at 500 IU/ kg dossage.Hepatic tissue concentrations of MCP_1, TGF_B and NF_KB, serum liver enzymes and lipids and hepatic tissue histological and structural changes were determind.Results: Vitamin D administration significantly reduced the MCP-1 concentration in HFD+vitamin D compared with HFD group and reduced liver TGF_B in both vitamin D treted groups ( p<0.05). Moreover, a significant reduction in the serum values of AST and ALT in vitamin D treated groups and in serum TG and TC in HFD+ vitamin D group. A pronounced improve in the markers of histological damage were also identified.Conclusion: Our data presented the obesity_induced inflammation and metabolic and histological abnormalities in the liver of high fat diet induced rats. We also demonstrated the potential benefit of vitamin D in ameliorating the inflammatory, metabolic and histologic changes in the liver of these animals.
