Background and Aim: Polycystic ovary syndrome (PCOs) is the most common cause of infertility in women. The risk of type-2 diabetes mellitus (T2DM), hyperinsulinemia and insulin resistance is higher among women with PCOs. We aimed to evaluate the effect of psyllium supplementation on insulin resistance and the lipid profile in non-diabetic (selection of non-diabetic subjects due to the adjustment of the effect of reducing blood glucose drugs) women with PCOs.Methods: A randomized double blind placebo-controlled trial. 54 eligible non-diabetic women with PCOs aged 18-45 recruited from an endocrinology clinic were divided into two groups based on their BMI through Stratified-Block Randomization. Participants in the intervention and placebo groups received 5g of psyllium or cellulose microcrystalline respectively twice a day for 8 weeks. Anthropometric indicators such as weight, BMI, WHR, fat mass and fat free mass, fasting insulin, FBS, HOMA-IR and several important indexes of lipid profiles were evaluated before and after the interventionResults: In the psyllium group the FBS, fasting insulin, total cholesterol, LDL-C, HOMA-IR decreased significantly (P<0.05) after 8 weeks, but the changes in these indexes were not significant in the placebo group. The results showed a significant decrease (P <0.05) in mean change of LDL-cholesterol in intervention group compared to placebo groupConclusion: Psyllium supplementation decreased FBS, fasting insulin, total cholesterol and LDL-C and improved insulin resistance in non-diabetic women with PCOs.
