THE EFFECT OF GENISTEIN ON INSULIN RESISTANCE IN COMPARISON WITH LETROZOLE INDUCED POLYCYSTIC OVARY SYNDROME RATS

Background and Aim: Polycystic ovary syndrome (PCOS) is one of the most prevalent gynecologic diseases in the world. Women with PCOS suffer from hyperandrogenism irregular menstruation, hirsutism and more importantly insulin resistance (IR). Genistein as one of major isoflavones showed favorable results on insulin action and glycemic status. The aim of this study was to compare the effectiveness of Genistein supplementation and metformin on glycemic status and some related indices in PCOS induced rats.Methods: PCOS was induced by 1 mg/kg letrozol in adult Sprague-dawley rats for 21 days and after that for 42 days, rats were treated by normal saline (NC), 150 mg/kg of metformin (MET) or 20 mg/KG of genistein dissolved in 1% methyl-cellulose solution (G). Body weight, glycemic and lipid profile, inflammatory and oxidative parameters as well as histopathological evidences were assessed at the end of intervention.Results: Despite slightly lower weight gain in G group during the study, no significant different observed between NC, MET and G groups. Treatment with metformin and Genistein significantly alleviate the increased level of fasting blood insulin (p=0.002), (p=0.16) and HOMA-IR (p<0.001), (p=0.012) however, only metformin group experienced decrease in fasting blood glucose (p=0.003). In addition, only G group had significantly lower level of serum malondialdehyde (p=0.039) and tumor necrosis factor-alpha (p=0.003) and higher activity of superoxide dismutase enzyme activity (p<0.001). Histological data showed that metformin and genistein therapy led to cysts disappearance and forming healthy follicles.Conclusion: Analysis of biochemical and histological evidences shows that genistein may be beneficial in PCOS not any less than metformin.
