Induction of atherosclerosis related micro RNAs byHelicobacter pylori and Lactobacillus acidophilusin effector memory T cells of patients with abdominal aortic aneurysm

Introduction and Objectives:Apoptosis and inflammation are two major players in abdominal-aortic-aneurysm (AAA). So, we evaluated Helicobacter pylori (Hp) and Lactobacillus acidophilus (La) mediated apoptosis and miRNA expression in human-umbilical-vein endothelial cells (HUVEC) and CD4+ effector-memory-T (Tem) cells of patients with AAA.Materials and Methods:MiR-21, 92a, 146a, 155, 326 and 663 were measured in sera and tissue of 8 AAA patients and in 8 controls sera. HUVECs and Tems separately and in co-culture were treated with CagA-positive/negative Hp-water-extract (HpWE) and La-conditioned-medium (LaCM). Apoptosis and miRs were evaluated using flowcytometry and real-time-PCR.Results:Positive correlations existed between apoptosis and miR-92a, 155, 326 and 663 in sera. MiR-155 was higher in patients than controls, while miR-155 and miR-146a were higher in the aneurysm tissue than serum. In single-culture, HpWE increased and LaCM decreased apoptosis in HUVECs and Tems similarly in patients and controls. HpWE increased miR-155 and LaCM increased miR-21 in Tems. In co-culture, HpWE increased apoptosis in Tems and LaCM modulated the effect of CagA+HpWE; LaCM induced more apoptosis in HUVECs and decreased miR-155 and miR-663 in Tems.Conclusion:Regardless of patients and controlsit seems that, H. pylori triggering apoptosis and pro-atherogenic miRNAs expression in CD4+Tem and ECs can participate in the initiation of AAA while La with apposite effects can prevent it, but at the late stages of the disease both microorganisms seem to be able to enhance the AAA. The differential effect of Hp and La on some atherosclerosis related miRNAs may be useful in future preventative approaches.
