The frequency of HPV genotypes in different grade of cervical lesions, in Fars province, 2016

Introduction and Objectives:Cervical cancer is the fourth prevalent cancer among women worldwide. HPVs frequently detected in almost all of cervical cancer lesion. Although, HPV 16149and 18 are the two most prevalent high risk papilloma virus types, but other genotype with different frequency might be detected in cervical lesion. Therefore, the aim of this study was to investigate the frequency of HPV genotypes in different grade of cervical lesions.Material and methods:Total of 103 FFPE samples with histopathologic diagnoses of cervical intraepithelial neoplasia and cervical cancer were included. DNA was extracted and presence of HPV16-18 was investigated by using Taq-man probes. For detection of other genotypes, nested PCR test with HPV general MY09/11 and GP5/6 primers runs for negative HPV16/18 samples and PCR products sent for sequencing.Result:The mean age of participants‟ was 43±1. After Taq-man real-time PCR, 80 samples were positive for HPV 16 or 18. Among them, 49 samples were HPV 16-18 co-infection, 21 samples were HPV 16 and 10 samples HPV18 single infection. Sequencing and alignment of PCR product of negative HPV 16 and 18 samples also showed that 2, 3, 1, 1 and 4 samples were infected with HPV genotype 6, 31, 51, 53 and 16, respectively. Conclusion:According to our results, the most common HPV genotypes were HPV 16, 18. Other genotype such as genotype 31, 6, 51 as well as 53 might be detected and involve in cervical cancer pathogenesis. This issue should be noted in HPV prevention program by HPV vaccine.
