Detection of Brucella Abortus in Kerman dairy cattle by Real-time PCR

Brucellosis is a major public health problem in many countries in the Middle East, such as Iran. In developing countries, milk contamination with Brucella is one of the most important ways of transmitting of the microorganism to humans. In order to detect brucellosis, the isolation of Brucella genus is one of the main methods of diagnosis, but for this purpose, equipment with a high biological safety level and a long time is required. To overcome these problems, methods with high sensitivity and high specificity, such as polymerase chain reaction, are commonly used. Among the various types of PCR, the Real Time PCR method is considered as a very suitable method. The aim of this study was to detect the bacteria of Brucella abortus in bulk tank milk of dairy cattle farms in the Kerman prvince using Real Time PCR technique. In this cross-sectional study, which was conducted from spring to autumn of 2017, 48 milk dairy herds were collected from 4200 milking cows. After transferring of the milk samples to microbiology laboratory, DNA was extracted by lysozyme buffer and proteininase K. Then, Specific primers wer used to detect fbaA gene belonging to Brucella abortus by Real Time PCR. The samples containing 176 bp fragment were considerd as Brucella abortus-positive samples. The results of this study showed that 3 milk samples (25.26%) were positive for presence of Brucella abortus. Therefore, Brucella abortus is present in the dairy cattle herds in Kerman, which indicate the importance of administration of preventive strategies.
