Anti-tumor property of hydroalcoholic extract of Oleo-gum-resin of Ferula assa-foetida on Gastric Cancer MKN-45Cells

Background and Objective: Gastric cancer is the fourth leading cause of cancer-related death in the world with a highest rate of mortality in the East, South and Central Asia as well as South America. It has been estimated that about 50% of all cancers are gastric in Iran. Despite recent improvement in cancer therapy protocols, such as surgery, multi-therapeutic agents and ionizing radiation, but many cancers remain untreatable due to its resistance nature or some side effect of the protocols. So, many researchers around the world attempt to find new strategies for solving the above mentioned problems. Among these novel strategies using herbal plant extracts that exert antitumor activity have received more interest as an alternative cancer therapy method because of less side effects and cost effectiveness. Ferula assa-foetida, one of 130 species of family Apiaceae, distributed throughout the Mediterranean and Central Asia. It also found in different regions of Iran, Southern Khorasan, Sistan and Baluchestan, and Kerman. Oleo-gum-resin (Asafoetida) obtained from the stems and roots of Ferula plants contains a number of bioactive compounds, including galbanic acid, farnesiferol, ferulic acid and its esters, coumarins, umbelliferoneand etc. Asafoetida is an effective remedy for several upset stomach, and studies have also shown its anti-cancer effects on some cancer models but till now its effect was not evaluated on gastric cancer one. So, in the present study, we decided to evaluate its antitumor activity on gastric cancer MKN-45 cell line. Material and Methods: For this aim, oleo-gum-resin of Assa-foetida was collected from Southern Khorasan, Tabas region, and its hydroalcoholic extract was prepared and freeze-dried. MKN-45 cells were cultured and treated with the extract at different concentrations range from 500-1000 ug/ml for 24h and 48h. Morphological changes in treated cells were evaluated using digital camera. Cell toxicity was carried out using MTT assay. Cell distribution at different stage of cell cycle was determined using PI-staining by flow cytometry and the percent of apoptotic cells were determined as sub-G1 population. Findings: Microscopic images showed morphological changes in treated MKN-45 cells as decrease in cell density and floating dead cells. MTT results demonstrated increase in cell toxicity in a time and dose dependent manner with IC50 of 800 ug/ml at 24h. Cell cycleanalysis of MKN45 cells treated with ferula extract (1000 ug/ml) showed significant cell arrest at G2/M stage. Cancer cell death was happened mostly in apoptosis manner by 24% at 800 ug/ml compare to control cells at 24h. Conclusion: Our results demonstrated that Assafoetida has a cytotoxic activity and antitumor effects against humangastric cancer MKN-45 cells. It seems that Assafoetida has a set of natural compounds that have anticancer effects.
