چکیده مقاله:

Background: HCV virus abuse the autophagy to increase its replication in host cell. The study crosstalk between Autophagy and HCV virus will lead to clear the HCV virus interaction with host cells in detail. The goal of this study was to essay the effect of autophagy induction after virus infection in Huh7.5 cells. Material and Method: To this end, the Huh7.5 cells were cultured, and theninfected with HCV (JFH1) virus. After 12 hours the recombinant pcDNA-Bclin 1 was transfected. Finally, the virus RNA was extracted and viral load was measured using Real-Time PCR 48hours post infection. Result: The result of this study suggests that the autophagy induction after virus infection can increase virus yield up to 3 fold in comparison with control group. Conclusion: Inhibition of autophagy after virus infection in Huh7.5 cells would be a novel target to design new antiviral compounds. In addition, it paves the way to better understand the fate of virus and host cell after the virus infection.
