Analysis of two allelic HLA haplotypes in different ethnicities of Khuzestan Province

Introduction: There is accumulating evidence that the HLA gene variants are associated with diseases. This surveyon population included a number of the haplotype frequencies in Khuzestan province. HLA allele recognition ishelpful in anthropological, transplantation and disease studies. Therefore, the aim of this study was investigatingthe association of HLA-DQA1*0102, -DQB1*0602, -DRB5*01 and -A*0301 haplotypes in normal populationaccording to Arab and Persian ethnicity.Methods: HLA-DQA1*0102, DQB1*0602, DRB5*01 and A*0301 genotypes were examined in Iranian normalpopulation of Khuzestan analyzing by polymerase chain reaction using specific sequence primers (PCR-SSP)method in 242 healthy blood donors without any disease. We studied the relationship of genotypes with ethnics.The statistical analyses were performed by SPSS software.Result: It was found that DQA1*0102+-DRB5*01- haplotype was associated with Persian ethnic compare withArabs (71% vs. 54.2%, p = 0.029, OR=0.481[95% CI =0.248-0.933]). However, it was not found any significantassociation between both Arab and Persian and following two allelic haplotypes: A*301- -DRB5*01- (58.13% vs65.9%, p =0.467, OR=0.720 [95% CI =0.297- 1.745]), DQB1*0602+-DRB5*01- (35.5% vs 46.6%, p=0.170,OR=0.632 [95% CI =0.328-1.220]). DQB1*0602+-A*301- (44.5 vs 50%, p=0.604, OR=0.800 [95% CI =0.344-1.860]).Conclusion: This study gives an insight into the HLA genetic pattern of normal population in Khuzestan province.It was shown that the DQA1*0102+-DRB5*01- haplotype has association with ethnicity in this population. AsKhuzestan is a high risk region for some diseases; thus, these findings encourage future research to investigate theinfluence of this haplotype in pathogenesis of the diseases.
