Introduction&Aim: Breast cancer is the most common cancer among women in the world, but its diagnosis in the early stages increases the chance of recovery and survival of the patient. In this study,we investigated the methylation of GBP2 and AKAP4 genes as a breast cancer biomarker in tumor tissue and peripheral blood in patients with breast cancer.Methods: In this study, possible changes of GBP2 and AKAP4 promoter methylation pattern in tumoral tissues and peripheral blood in comparison with normal ones, in presence of a house keeping gene as an internal control in 60 breast cancer patients was analyzed using MS-PCR technique.Results: At this study we found genes that were up regulated and down regulated in breast tumor. AKAP4 and GBP2 were down regulated for cause of methylation modification. GBP2 as a tumor suppressor gene was hyper methylated and down regulated in breast tumor samples. AKAP4 as a cancer testis antigen was hypo methylated and down regulated as well in breast tumor samples.Conclusion: As a consequence, GBP2 and AKAP4 promoter methylation pattern are associated with breast cancer as possible biomarkers. Also GBP2 and AKAP4 promoter methylation pattern may influence their expression and affect cancer pathologic characteristics. More information in this field may be promising in cancer management.
