The inhibitory effects of hesperetin on MCL-1, BCL-XL, and BCL-2 anti-apoptotic factors in PC3 cancer cell line

چکیده

Introduction & Aim: Hesperetin is an antioxidant exists in variety of plants. Nowadays, some of antioxidants can induce the apoptosis in some cancer cells by activation of apoptosis factors. In this study, we evaluated the effects of hesperetin on MCL-1, BCL-XL, and BCL-2 anti-apoptotic factors in PC3cancer cells.Methods: Cell viability was assessed by MTT assay. Comet assay and flow cytometry technique were done at three different concentrations below IC50 of hesperetin. 100 comet pictures have been analyzed with CASP V:1.2.2 software and then all results were analyzed by SPSS V:21 statistical software. Simulation studies were done by Auto Dock, VMD and Gromacs soft wares on evaluation MCL-1, BCL-XL, and BCL- 2 anti-apoptotic factors Results: Hesperetin led to induce apoptosis in PC3 cells. The IC50 value of hesperetin was calculated at450 μM. The ratio of tail DNA to head DNA in comet assay was 44.5±1.8 and 24.7±2.7 for 450 and 400 μM hesperetin, respectively. In simulation and molecular dynamic studies, the lowest binding energy between the hesperetin and MCL-1, BCL-XL, BCL-2 were -6.70, -7.05, and -6.73, respectively. Conclusion: Hesperetin not only led to genome demolition of PC3 cancer cells but also resulted in the induction of apoptosis via decrease MCL-1, BCL-XL, and BCL-2 anti-apoptotic factors
