In vitro production and cytotoxic effect of Withaferin A on Esophageal Cancer cell line (KYSE30)

The genus Withania (fam. Solanaceae) comprises 26 species. This genus in the flora of Iran is represented by 2 species W. somnifera and W.coagulans, both of which have high medicinal value. The main medicinal activity of this genus related to present of steroidal lactones, known as withanolides. The most important of which is the potential anticancer compound withaferin A, that recently has gained much attention in medicinal industry. In the present study, In vitro production of Withaferin A by Hairy root culture and elicitation by Aspergillus niger, Alternaria alternate and Fusarium solani extract was accomplished. Finally, the cytotoxicity effect of extract (0-16 µg/ml) on Esophageal Cancer cell line (KYSE-30) was evaluated by MTT test. The successful hairy root induction was achieved by using A. rhizogenesis strain A13 on ½ MS medium. The highest amount of Withaferin A (168.16±4.39 µg/g D.W) was produced in hairy root by elicitation with Aspergillus niger extract (5% V/V). Hairy root extract showed cytotoxicity effect on KYSE-30 cancer cells (IC50; 21µg/ml). In conclusion, this study has found that generally root extract has more cytotoxic effect on KYSE30 than noncancerous HFF3 cells. In conclusion, however, development and implementation of effective programs to conservation, reclamation and efficient utilization of valuable plant species seem to have a special importance.
