Staphylococcus epidermidis is a main cause of nosocomial bacteremia, prosthetic valves endocarditis, surgical wounds, central nervous system shunt infection, infections related to intravenous catheters, infections related to dialysis, and artificial joint infection. Therefore identifying this bacterium is very important. Phenotypic identification of coagulase-negative staphylococci species are not satisfying and reliable. Thus, PCR-based methods to identify these species have been investigated. The aim of this study was to design a PCR technique for the pta gene in order to identify bacteria Staphylococcus epidermidis. The pta is a housekeeping gene that plays a role in the metabolism of acetate. The primers were designed for pta gene. Staphylococcus epidermidis ATCC 12228 was obtained from the Pasteur Institute and cultured on the suitable medium. Bacteria DNA was extracted by boiling manner. PCR with designed primers for pta gene leads to amplifying DNA of Staphylococcus epidermidis. The method used in this study to identify Staphylococcus epidermidis, is easy, fast and precise and can be used for more samples of Staphylococcus epidermidis to study specificity of the pta gene accurately.
